Why An Autosampler Is A Vital Partner For HPLC Analysis

Chromotography is the science of splitting compounds into their constituent chemicals and is achievable via a variety of methods using many different instruments. To achieve its main goals HPLC will use an autosampler to simplify the process. Let us take a closer look at how these instruments work closely together.

Column chromatography is a method of separating compounds which drives a solvent through a column having a small sample of the compound within. Based on specific qualities of how the solvent interacts with various constituent components of the compound, some will be separated out of the original mixture while others will remain. This is basically how HPLC is developed.

HPLC doesn’t allow the solvent to seap through the column as it does with column chromatography. Rather, the solvent is forced through at much greater pressures. The advantages are two-fold.

First, because the pressures are much greater, often as high as 400 atmospheres, the process is dramatically quicker. Were the solvent simply dripping through under the pressure of gravity, separating the compound into its component elements would be a much longer process.

Also, materials on a smaller scale can be used as the packing surface within the column. The result is a much more accurate separation of the desired components that would be possible via traditional column techniques.

However, these advantages do come at a cost. Other machinery is needed to carry out the extreme pressures required on the solvent. Also, the more detailed separation of the desired components needs a greater level of detection. This is when an autosampler is used. Generally, HPLC autosampler products perform these main steps.

First of all the sample is injected into the main column. At this point high pressures are produced to push the compound through the column at very high velocities.

The particles go through the column via the solvent and then {finally reach the detector}. working out the separation is quite simple by measuring the time it eakes for individual chemicals to extend the column~If we know the sizes of the column and solvent, as well as the main pressure we can determine the time taken for the separation of the compound and the time it takes the substance to pass through the column}.

This value, known as the retention time, is one of many factors used by the detector in determining when a given chemical has been removed. Using retention time as a detection method also requires careful temperature control, as temperature affects the flow rate of compounds through the column using retention time as a method of detection}. This is just another main factor maintained by HPLC autosampler products.

When the compound moves through the column, a detection process occurs by using a number of different methods. One popular method is the use of UV light and an ultraviolet detector. By shining UV light through the stream and detecting which wavelengths are absorbed, an accurate identification
of some chemical compounds can be obtained.

The result of ultra violet spectroscopy, tetention timings and other detection is then translated by computers to form an identity of the components chemical properties.

It is almost impossible to perform HPLC operations manually due to its high pressures and vast speed. As such, an autosampler is an invaluable tool when using this method of chromatography.
